KPV is about as small as a peptide gets. It is three amino acids, lysine, proline, and valine, snipped from the tail end of a hormone your body already makes called alpha-MSH. That parent hormone is one of the natural brakes on inflammation, and KPV appears to keep most of that braking ability while dropping the effects on skin pigment that alpha-MSH also carries.
The interest in KPV is mostly about the gut. Unlike most peptides, it survives being swallowed. Intestinal cells pull it in through a transporter called PepT1, which means an oral dose can act directly on inflamed gut tissue rather than needing an injection.
Once inside the cell, it interferes with NF-kB, a master switch that turns on inflammatory genes. Turn that switch down and the output of inflammatory messengers like TNF-alpha and IL-6 drops with it.
It is also used topically for inflammatory skin conditions, on the same anti-inflammatory logic. None of these uses are approved anywhere, and the human evidence is essentially absent.

The anchor study is a 2008 paper in Gastroenterology showing that oral KPV reduced intestinal inflammation in two separate mouse models of colitis. It established both the mechanism, uptake through PepT1 followed by NF-kB inhibition, and the practical point that swallowing it works.
Follow-up work in cells and animals has been consistent. KPV reduces inflammatory signalling, supports the mucosal barrier, and helps restore tight junctions between the cells lining the intestine. Some of this work has explored delivering it in nanoparticles to concentrate it where inflammation is worst.
The gap is human data. No completed randomized trial has tested KPV on its own in people. Everything you read about it in the context of IBD, IBS, or leaky gut is an extrapolation from mouse studies.
Its regulatory position in the United States has been unsettled. It was removed from the restricted compounding list in April 2026 after the nomination behind it was withdrawn, and related substances were scheduled for further review that July. This is worth watching if you follow the compounding rules, but it says nothing about whether the peptide works.
